Probably Zoe or Wash. Zoe certainly served well as the "kick-butt female" role, but she was never very deep or developed as a character. Wash was also a rather 2-D character and while he was funny, his quips were starting to annoy me by the last couple episodes. He was kind of like Xander in that way, but amused me less than Xander did.
In terms of "what might have been", I think that it'd be Jayne or Inara (although I don't actually DISLIKE either of them) While I like Jayne, I just feel he'd quickly become just the "brute". Whatever development he has can't be great, as that would no longer fit in terms of his character (i.e. he wouldn't be Jayne anymore). To me, he's a bit like Spike, without the extra personality/looks to make him more entertaining. Like Spike, the only real area for "growth" he has while remaining the same character is merely increased ethical development. Inara, actually, I think has some of the least room for development as a character, BUT, I think Joss had forseen that and that was part of the reason why she left the ship. Given that, and how I find her amusing and sexy, I think I'd probably put Jayne a bit more "disliked", but not much.
For the record, I think that the character of River did have a great deal of potential if Joss chose to work with it. I think the biggest "danger" of River's character would have been continuing to use her primarily has a plot device instead of an actual persona. The characters of both Kaylee and Simon were simple, naieve and "raw" enough to have a lot of potential (much like Willow developed a lot throughout Buffy). I'm not sure exactly where Mal had the future of going. I could equally see a lot of development being put into him, or a lot of stagnation at some point. Like Zoe, he is a more "complete" character and those are always more difficult to develop than a character like Simon or Kaylee.
I absolutely adored Shepard and I think they could have done a lot with him as a character. His development would be more "backstory revelation" oriented, much like Giles was developed. That's what you have to do with older "complete" characters, though.
And I've put way too much thought into this, I think...
