If the game doesn't support gamepads, you'll have to use software to rebind your keyboard to the controller.
Either way, though, a wired 360 controller is USB and runs perfectly with a PC. I use one to play my emulated N64 games.

Certain games, often those from XBLA, will ONLY support the 360 pad for the controller option.
