Super Mario Galaxy is the most tightly designed 3D platformer ever made, bar none. If you have any interest in platformers at all, try it out. If you're one of those who loved the old 2D Marios but didn't like the 3D ones, try it out. The structure of this one, where you go from the beginning of the level to the end a opposed to exploring a space, matches the 2D games.

Twilight Princess is great.

Super Smash Bros Brawl is amazing.

Okami is great, but it's better on the PS2, so only get it for the Wii if you don't have a PS2.

Super Paper Mario is an interesting cross between RPG and platformer, with a cool gimmick and hilarious writing.

Megaman 9 is great is you're nostalgic about old Megaman games, otherwise probably avoid it.

Muramasa: The Demon Blade is a great sidescrolling beat-em-up type game with absolutely gorgeous art style.

Lost Winds is a fun, side scrolling adventure game with a bit of a Zelda vibe, and it makes good use of the wii remote.

Resident Evil 4 is good as ever, but it's available on some other systems, so I don'tr know which is best.

Metroid Prime trilogy isn't really my thing, but I know it's loved by many.

New Super Mario Bros: it's like Super Mario Bros, but with ridiculous 4-player party action.

And then there's a lot of good, but niche games: Monster Hunter Tri, Sin and Punishment, Zack and Wiki, De Blob, etc. These are ones that some people will love, many people won't, so I can't necessarily recommend them without knowing your tastes better.
